Sophia é filha de um sacerdote da Igreja Anglicana e de uma publicitária. Foi aceita na Universidade de Cambridge, é fluente em três línguas (inglês, alemão e francês) e aprecia montar cavalos, esquiar e nadar. Um diretor de casting da BBC a descobriu em uma peça de teatro de seu colégio aos dezesseis anos de idade e a convidou para a versão televisiva de O Príncipe e o Mendigo.
Ela também atuou na minissérie Big Women, no seriado Mansfield Park e em uma adaptação para a televisão da biografia de Oliver Twist.

Myles estava em um relacionamento com o ator Charles Dance, que é 34 anos mais velho que ela, que ela conheceu enquanto trabalhava em A Vida e Aventuras deNicholas Nickleby.
De 2005 a 2007 ela namorou o ator escocês David Tennant, com quem ela tinha agidode guerra de Foyle e Doctor Who episódio "The Girl in the Fireplace".
